SARS-CoV-2 Products Affinity Test
Bio-layer Interferometry (BLI) was employed to measure affinity between SARS-CoV-2 RBD-His protein (K1516) and SARS-COV-2 antibody K79d9. The affinity constant Kd is below 1uM, which indicates strong interactions between the two SARS-CoV-2 products.
SARS-CoV-2
Description
SARS-CoV-2 (Severe Acute Respiratory Syndrome Coronavirus 2), also known as 2019-nCoV (2019 Novel Coronavirus) is a positive-sense single-stranded RNA virus belongs to the family of coronaviruses. It is the seventh known coronavirus to infect people after 229E, NL63, OC43, HKU1, MERS-CoV, and the original SARS-CoV.
Structure
SARS-CoV-2 processes four structure proteins, including the spike (S), envelop (E), membrane (M) and nucleocapsid (N). Coronavirus entry into host cells is mediated by the transmembrane spike (S) glycoprotein. The S glycoprotein comprises two functional subunits responsible for binding to the host cell receptor (S1 subunit) and fusion of the viral and cellular membranes (S2 subunit). ACE2 could mediate SARS-CoV-2 S-mediated entry into host cells, establishing it as a functional receptor for SARS-CoV-2. The SARS-CoV-2 S engages human ACE2 (hACE2) with comparable affinity to SARS-CoV S. SARS-CoV-2 is unique among known betacoronaviruses in its incorporation of a polybasic cleavage site, which increases SARS-CoV-2 pathogenicity and transmissibility in other viruses.
